Description

S-fil works by inhibiting the enzyme Phosphodiesterase type 5 (PDE5). Inhibition of PDE5 results in the accumulation of cyclic GMP (cGMP), a chemical that relaxes the smooth muscle walls of blood vessels (vasodilation).

  • For Erectile Dysfunction: During sexual stimulation, this vasodilation increases blood flow to the penis, which is necessary to achieve an erection. It is crucial to note that sexual stimulation is mandatory for S-fil to cause an erection.   
  • For Pulmonary Arterial Hypertension: It relaxes the pulmonary vasculature, reducing blood pressure in the lungs and decreasing strain on the heart.   

Dosage and Administration

S-fil should only be taken according to the specific directions provided by your doctor.   

  • Dosage: Senigra (50mg or 100mg) should not be taken more than once per day.
    • For ED: The usual starting dose is 50mg, taken as needed . Based on efficacy and tolerability, the dose may be increased to a maximum of 100mg or decreased to 25mg under the guidance of a physician .
  • Timing: Take the tablet approximately 30 minutes to 1 hour before sexual activity. The effect can last up to 4 hours.   
  • Food and Drug Interactions:
    • Taking S-fil with high-fat meals may delay the onset of action (peak plasma levels can be delayed from 30 minutes up to 120 minutes).   
    • Avoid Alcohol, as it may increase the risk of dizziness and difficulties achieving an erection.   
    • Grapefruit juice should be avoided as it can elevate S-fil levels in the body, increasing the risk of side effects.

Absolute Contraindications (Strictly Forbidden)

S-fil must NEVER be used in combination with the following, as the interaction can cause a life-threatening drop in blood pressure (Severe Hypotension):

  1. Nitrates: Medications used for chest pain (Angina), such as Nitroglycerin or Isosorbide Mononitrate, or recreational nitrates (“poppers”).   
  2. Guanylate Cyclase Stimulators: Medications like Riociguat or Vericiguat.   
  3. Allergy: Known hypersensitivity to S-fil or any component of the formulation.   
  4. Vision Issues: History of Non-arteritic Ischemic Optic Neuropathy (NAION) or inherited degenerative retinal disorders .

Serious Warnings (Immediate Medical Attention Required)

Stop taking Senigra and seek emergency medical care immediately if any of the following symptoms occur :   

  • Priapism (Prolonged Erection): An erection lasting longer than 4 hours, which can cause permanent damage to the penis if untreated.   
  • Sudden Sensory Loss: Sudden loss or decrease in hearing (sometimes with tinnitus and dizziness), or sudden loss of vision/blurred vision .
  • Cardiac Events: Chest pain, severe dizziness, fainting, or shortness of breath.   
  • Severe Allergic Reaction: Swelling of the face, lips, throat, or tongue; difficulty breathing; or severe rash/hives.

  • Dizziness (Do not drive or operate machinery if you feel dizzy).   
  • Changes in vision (e.g., a slight blue tinge to objects, sensitivity to light)

Less Common Side Effects

Less frequently reported effects include vomiting, sleep disturbances, muscle or back pain, rash/itching, and rapid heartbeat or low blood pressure.
